(1.If all cells have the same DNA, how do we make different cells and organs? 2.In your own words, define clone.  3.Describe what you know about the science of cloning.(For example: What do you know about the science of cloning genes??) 4.What are the ethical considerations surrounding cloning?)
2. Cloning Video- Day 1

H.W. Typed Transformation Lab Report (due TOMORROW)
Create the following headings and then place the requirements that are listed below after them:

  • Title: Title the lab
  • Purpose:  State the purpose of the lab in YOUR OWN WORDS
  • Background Information: Take the 10 bullet points that you wrote after reading the Transformation Lab Background Information and create 2 cohesive paragraphs.  Your bullet points should already be in your own words, but if they are not, make sure that the 2 paragraphs are.  You are paraphrasing : "a restatement of a text, passage, or work giving the meaning in another form."
  •  Prelab:  Type the answers to your prelab questions.
  • Hypotheses:  Type your 4 scientifically "meaty" hypotheses
  • Materials:  Read the Transformation Lab Procedure and bullet point all materials that were used in the lab.  Ex. 4 sterile inoculating loops, 4 pipettes, thermometer, beaker, ice, hot water bath etc.....
  • Procedure:  Type this heading.  Under the heading type "see procedural flow chart on page ____."  You will then attach your colorful procedural flow chart that you have already completed to your lab.
  • Data:   Type this heading.  Under the heading type "see data on page ____."   You will then attach your data drawings from today to your lab.  For each plate you need to draw (in color) and describe your data (ex. how many colonies, no growth, lawn growth etc...).  Each plate must also be labeled with what was on the plate (Ex. LB+, LB-, etc...)
  • Analysis:  Answer the following questions in complete, well thought out sentences. 
    1. On which plate(s) did you see transformed bacteria?  Explain, in detail why the transformed bacteria were successful in this medium.
    2. Which plate(s) had lawn growth?  Did the lawn growth also glow?  Explain, in detail why this type of growth occurred.
    3. What allowed you to isolate the transformed cells (those that took up plasmid) from those that did not take up the plasmid?
    4. If you did not get any results (transformation did not occur), what factors could have affected your experiment?*** only for groups that did not have successful transformation
    5. In a cohesive paragraph, describe how scientists create recombinant DNA and transform bacteria.  Also describe & give examples of how scientists apply this transformation technique to scientific research.
     
  • Attach your stamped rough draft to the back of your formal lab report.  Your rough draft should have a total of 2 stamps.

2. Transformation Lab: the exciting results!!!!  (absent today?  Make sure you get your lab results from another member of your lab group.  Also, on Monday I need to stamp your procedural flow chart and hypotheses)

H.W. Start typing your Transformation Lab.  This is due on TUESDAY.  Create the following headings and then place the requirements that are listed below after them:
  • Title: Title the lab
  • Purpose:  State the purpose of the lab in YOUR OWN WORDS

H.W. Finish your lab hypotheses (see below) and start typing your lab (more info to come tomorrow regarding typing your lab)
Transformation Lab Hypotheses: You will have a total of 4 hypotheses.
•For the E. Coli in each petri dish say if you expect growing AND glowing. These are the contents of your four plates:
a.LB/Amp+ (luria broth, ampicillin, plasmid) – this is an experimental plate
b.LB/Amp- (luria broth, ampicillin, no plasmid) – this is a negative control
c.LB+ (luria broth, no ampicillin, plasmid) – this is a positive control
d.LB- (luria broth, no ampicillin, no plasmid) – this is a positive control
•Use an if….then….because statement for each one of the plates above.
For example:
•If the petri dish contains _____________ (ex. Luria broth and Ampicillin) and the E.coli is given/not given (choose one) the plasmid, then the E.coli will ___________ (grow and glow, grow and not glow, not grow and not glow ect….) because ___________(insert something scientific here based on the background info)

1. Read the Transformation Lab Procedure and then draw a flow-chart style picture using color to depict all of the steps of the lab. Create a brief description to describe each picture/step.  This is to be completed on a white sheet of construction paper.
2. Transformation Lab- "dry run/ dress rehearsal"

H.W. Finish your procedural picture/flow chart (see instructions under #1 above).  You will work on your hypotheses tomorrow in class during the 30 minute recovery period.

Reminder:  Those students who did not complete the h.w. from last night must turn in their prelab assignment before 1st period in order to participate in the lab.  In other words: no prelab = a ZERO on the entire lab.

Part I:  Take out a new sheet of paper.  Title the paper "Transformation Lab."  Create a "Background Information" heading.  Read the Transformation Lab Background Information (click on the name of the document to open).  Then paraphrase (use your OWN words) 2-4 important bullet points from each paragraph under this heading.
Part II:  Create a "Prelab Questions" heading.  Read the Transformation Lab Procedure and then answer the following questions in complete sentences under this heading.
  1. What does it mean when a cell is competent? 
  2. How do we make cells competent?
  3. What are two important sequences on a plasmid? Why are they important? 
  4. Describe our gene of interest (What is the source? What protein does it code for?). 
  5. How many tubes will you have? What goes in each tube? 
  6. How many plates will you have? Draw 4 plates (circles).  Under each plate describe what  will go on each plate?
